Dysregulated Expression in Human Cancer Platelet-derived Growth Factor D : Tumorigenicity in Mice and Updated Version
نویسندگان
چکیده
Platelet-derived growth factor (PDGF) has been directly implicated in developmental and physiological processes, as well as in human cancer and other proliferative disorders. We have recently isolated and characterized a novel protease-activated member of the PDGF family, PDGF D. PDGF D has been shown to be proliferative for cells of mesenchymal origin, signaling through PDGF receptors. Comprehensive and systematic PDGF D transcript analysis revealed expression in many cell lines derived from ovarian, renal, and lung cancers, as well as from astrocytomas and medulloblastomas. PDGF receptor profiling further suggested autocrine signaling in several brain tumor cell lines. PDGF D transforming ability and tumor formation in SCID mice was further demonstrated. Exploiting a sensitive PDGF D sandwich ELISA using fully human monoclonal antibodies, PDGF D was detected at elevated levels in the sera of ovarian, renal, lung, and brain cancer patients. Immunohistochemical analysis confirmed PDGF D localization to ovarian and lung tumor tissues. Together, these data demonstrate that PDGF D plays a role in certain human cancers.

متن کاملComparison of Effects of High-Fat, High-Protein and Low-Calorie Diets on Klotho Gene Expression and TGF-β Level in Serum and Liver of Old Male Wistar Rats
Background and Objectives: Different diets have important roles in human health and aging. In the current study, we aimed to characterize the Klotho, an anti-aging protein, gene expression and protein level in the serum and assess the protein levels of Transforming growth factor-β and Platelet-derived growth factor in old rats which treated with different diets (high-fat, high-protein, low calo...
متن کاملMalignant transformation of p53-deficient astrocytes is modulated by environmental cues in vitro.
The early incidence of p53 mutation in astrocytomas suggests that it plays an important role in astrocyte transformation. Astrocytes isolated from homozygous p53 knockout mice grow rapidly, lack contact inhibition, and are immortal. Here we tested whether the loss of p53 is sufficient for progression to tumorigenicity of astrocytes. We grew primary astrocytes under three conditions for over 120...
متن کاملTumorigenicity of Esophageal Cancer Stem Cells (ECSCs) in nude mouse xenograft model
Background and objectives: Modeling cancer in vivo is a very important tool to investigate cancer pathogenesis and molecular mechanisms involved in cancer progression. Laboratory mice are the most common animal used for rebuilding human cancer in vivo. Cancer stem cells (CSCs) are the main reason of failure in cancer therapy because of tumor relapse and metastasis. Isolation of cancer stem cell...
متن کاملCritically Regulates the Tumorigenicity of Glioma Cells FoxM1B Is Overexpressed in Human Glioblastomas and Updated Version
The transcription factor Forkhead box M1 (FoxM1) is overexpressed in malignant glioma. However, the functional importance of this factor in human glioma is not known. In the present study, we found that FoxM1B was the predominant FoxM1 isoform expressed in human glioma but not in normal brain tissue. The level of FoxM1 protein expression in human glioma tissues was directly correlated with the ...
